Design of a Cyclodextrin Bioproduction Process Using Bacillus pseudofirmus and Paenibacillus macerans

Cyclodextrin (CD) drug delivery systems offer the potential to enhance desired physicochemical properties and pharmacokinetic parameters of drugs while maintaining their safety. Cyclodextrin-glucosyl-transferase (CGTase) is amongst most important enzymes used in CD biosynthesis. However, bioproduction CDs still faces challenges terms optimization process complexity. This study proposes a novel system batch mode increase yield reduce costs. Two bacterial strains were selected: alkalophilic Bacillus pseudofirmus DSM2517 strain neutrophilic Paenibacillus macerans DSM1574 strain. Three different culture media, two temperatures (30 °C 37 °C), three scales (shake flasks 20 mL 100 mL, bioreactor 3.2 L) evaluated with respect growth kinetics, protein production, CGTase biosynthesis activity for β-CD production. Bacterial was monitored by measuring optical density (OD600 nm), assessed production directly medium after filtration or samples concentration (using Vivaspin 500® ultrafiltration spin column 10 kDa cut-off). quantification performed using phenolphthalein colorimetric method HPLC. The best conditions combined both microorganisms, shake achieved containing 2% dextrin as carbohydrate source. Scale-up displayed improved kinetics bacteria higher macerans.
